Abstract
-
Obesity and metabolic dysfunction-associated fatty liver disease (MAFLD) are prevalent metabolic disorders with substantial global health implications that are often inadequately addressed by current treatments and may have side effects.
Probiotics have emerged as promising therapeutic agents owing to their beneficial effects on gut health and metabolism. This study investigated the synergistic effects of a probiotic combination of BNR17 and ABF21069 on obesity and MAFLD in C57BL/6 mice fed a high-sucrose diet. The probiotic combination significantly reduced body weight and fat accumulation compared with the high-sucrose diet. It also alleviated elevated serum leptin levels induced by a high-sucrose diet.
Histological analysis revealed a significant reduction in white adipose tissue and fatty liver in the mice treated with the probiotic combination. Furthermore, increased expression of genes related to β-oxidation, thermogenesis, and lipolysis suggested enhanced metabolic activity. The probiotic groups, particularly the BNR17 group, showed an increase in fecal exopolysaccharides, along with a tendency toward a lower expression of intestinal sugar transport genes, indicating reduced sugar absorption. Additionally, inflammatory markers in the liver tissue exhibited lower expression in the ABF21069 group than in the HSD group. Despite each strain in the combination group having distinct characteristics and functions, their combined effect demonstrated synergy in mitigating obesity and MAFLD, likely through the modulation of fecal exopolysaccharides content and improvement in lipid metabolism. These findings underscore the potential of probiotic supplementation as a promising assistant therapy for managing obesity and MAFLD and provide valuable insights into its therapeutic mechanisms in metabolic disorders.

Lipopolysaccharide (LPS) is a critical component of the extracellular leaflet within the bacterial outer membrane, forming an effective physical barrier against environmental threats in Gram-negative bacteria. After LPS is synthesized and matured in the bacterial cytoplasm and the inner membrane (IM), LPS is inserted into the outer membrane (OM) through the ATP-driven LPS transport (Lpt) pathway, which is an energy-intensive process. A trans-envelope complex that contains seven Lpt proteins (LptA-LptG) is crucial for extracting LPS from the IM and transporting it across the periplasm to the OM. The last step in LPS transport involves the mediation of the LptDE complex, facilitating the insertion of LPS into the outer leaflet of the OM. As the Lpt system plays an essential role in maintaining the impermeability of the OM via LPS decoration, the interactions between these interconnected subunits, which are meticulously regulated, may be potential targets for the development of new antibiotics to combat multidrug-resistant Gram-negative bacteria. In this review, we aimed to provide an overview of current research concerning the structural interactions within the Lpt system and their implications to clarify the function and regulation of LPS transport in the overall process of OM biogenesis.
Additionally, we explored studies on the development of therapeutic inhibitors of LPS transport, the factors that limit success, and future prospects.

Abstract

Bacteriophages (phages) are natural antibiotics and biological nanoparticles, whose application is significantly boosted by
recent advances of synthetic biology tools. Designer phages are synthetic phages created by genome engineering in a way
to increase the benefits or decrease the drawbacks of natural phages. Here we report the development of a straightforward
genome engineering method to efficiently obtain engineered phages in a model bacterial pathogen, Pseudomonas aeruginosa.
This was achieved by eliminating the wild type phages based on the Streptococcus pyogenes Cas9 (SpCas9) and facilitating
the recombinant generation based on the Red recombination system of the coliphage λ (λRed). The producer (PD) cells of
P. aeruginosa strain PAO1 was created by miniTn7-based chromosomal integration of the genes for SpCas9 and λRed under
an inducible promoter. To validate the efficiency of the recombinant generation, we created the fluorescent phages from a
temperate phage MP29. A plasmid bearing the single guide RNA (sgRNA) gene for selectively targeting the wild type gp35
gene and the editing template for tagging the Gp35 with superfolder green fluorescent protein (sfGFP) was introduced into
the PD cells by electroporation. We found that the targeting efficiency was affected by the position and number of sgRNA.
The fluorescent phage particles were efficiently recovered from the culture of the PD cells expressing dual sgRNA molecules.
This protocol can be used to create designer phages in P. aeruginosa for both application and research purposes.

Abstract

The use of antibiotics can disrupt the body’s natural balance and increase the susteptibility of patients towards fungal infections.
Candida albicans is a dimorphic opportunistic fungal pathogen with niches similar to those of bacteria. Our aim was
to study the interaction between this pathogen and bacteria to facilitate the control of C. albicans infection. Alpha-hemolysin
(Hla), a protein secreted from Staphylococcus aureus, causes cell wall damage and impedes the yeast–hyphae transition in
C. albicans. Mechanistically, Hla stimulation triggered the formation of reactive oxygen species that damaged the cell wall
and mitochondria of C. albicans. The cell cycle was arrested in the G0/G1 phase, CDC42 was downregulated, and Ywp1
was upregulated, disrupting yeast hyphae switching. Subsequently, hyphae development was inhibited. In mouse models,
C. albicans pretreated with Hla reduced the C. albicans burden in skin and vaginal mucosal infections, suggesting that S.
aureus Hla can inhibit hyphal development and reduce the pathogenicity of candidiasis in vivo.

Abstract

Tuberculosis, an infectious disease, is caused by Mycobacterium
tuberculosis. It remains a significant public health issue
around the globe, causing about 1.8 million deaths every year.
Drug-resistant M. tuberculosis, including multi-drug-resistant
(MDR), extremely-drug-resistant (XDR), and totally drugresistant
(TDR) M. tuberculosis, continues to be a threat to
public health. In the case of antibiotic-resistant tuberculosis,
the treatment effect of conventional antibiotics is low. Side
effects caused by high doses over a long period are causing
severe problems. To overcome these problems, there is an urgent
need to develop a new anti-tuberculosis drug that is different
from the existing compound-based antibiotics. Probiotics
are defined as live microorganisms conferring health
benefits. They can be potential therapeutic agents in this context
as the effectiveness of probiotics against different infectious
diseases has been well established. Here, we report that
Lactobacillus crispatus PMC201 shows a promising effect on
tuberculosis isolated from vaginal fluids of healthy Korean
women. Lactobacillus crispatus PMC201 reduced M. tuberculosis
H37Rv under co-culture conditions in broth and reduced
M. tuberculosis H37Rv and XDR M. tuberculosis in macrophages.
Lactobacillus crispatus PMC201 was not toxic to a
guinea pig model and did not induce dysbiosis in a human
intestinal microbial ecosystem simulator. Taken together, these
results
indicate that L. crispatus PMC201 can be a promising
alternative drug candidate in the current tuberculosis drug
regime. Further study is warranted to assess the in vivo efficacy
and confirm the mode of action of L. crispatus PMC201.

Paenibacillus polymyxa is a promising plant-growth-promoting
rhizobacterium that associates with a wide range of host
plants, including agronomically important ones. Inoculation
of wheat seedlings with P. polymyxa strains CCM 1465 and
92 was found to increase the mitotic index of the root cells
1.2- and 1.6-fold, respectively. Treatment of seedlings with
the exopolysaccharides (EPSs) of these strains increased the
mitotic index 1.9-fold (P. polymyxa CCM 1465) and 2.8-fold
(P. polymyxa 92). These increases indicate activation of cell
division in the root meristems. Analysis of the morphometric
variables of the seedlings showed that P. polymyxa CCM
1465, P. polymyxa 92, and their EPSs promoted wheat growth,
increasing root and shoot length up to 22% and root and
shoot dry weight up to 28%, as compared with the control.
In addition, both strains were found to intensely colonize the
seedling root surface. Thus, P. polymyxa EPSs are active metabolites
that, along with whole cells, are responsible for the
contact interactions of the bacteria with wheat roots and are
implicated in the induction of plant responses to these interactions.
The strains used in this work are of interest for
further study to broaden the existing understanding of the
mechanisms of plant–bacterial interactions and to develop
effective biofertilizers for agricultural purposes.

Abstract

Sanghuangporus sanghuang is a well-known pharmacodynamic
and economically important edible fungus associated with
mulberry (Morus spp.). A distinctly new exopolysaccharide
(EPS), designated SHP-2 was obtained from S. sanghuang
P0988 broth, and its structure and anti-aging prosperity were
characterized. SHP-2 was found to be composed of a backbone
of 4)-β-Manp-(14)-α-Araf-(13,4)-α-Glcp(1
3,4)-α-Glcp-(13,4)-α-Glcp-(13,4)-α-Glcp-(13,4)-α-
Glcp-(16)-α-Galp-(14)-β-Manp-(1and five branches,
including four α-D-Glcp-(1and one α-D-Manp-(1
SHP-2 was shown to increase antioxidant enzyme activities
including catalase (CAT) and superoxide dismutase (SOD)
activities, as well as trolox equivalent antioxidant (TEAC)
capacity in serum of mice pre-treated with D-Gal, while reducing
lipofuscin levels. SHP-2 exerted a favorable influence
on immune organ coefficients and ameliorated the histopathological
hepatic lesions and apoptosis in hepatocytes of Dgalactose-
aged mice almost in a dose-dependent manner.
Using the same analytical methods, on comparison with previously
studied EPS compounds (i.e. SHP-1), SHP-2 was found
to have more complex structure, larger molecule weight, and
different anti-aging properties. The results presented here suggest
that not only does EPS bioactivity vary with respect to
molecular structures and molecule weight, but that multiple
structures with different activity can be expressed by a single
fungal strain. These results may help understanding the antiaging
prosperity of these polysaccharides for use in health
foods or dietary supplements.

Abstract
-
Lipopolysaccharide (LPS) is one of the major components in
the outer membrane of Gram-negative bacteria. However,
its heterogeneity and variability in different bacteria and differentiation
conditions make it difficult to extract all of the
structural variants. We designed a solution to improve quality
and biological activity of LPS extracted from various bacteria
with different types of LPS, as compared to conventional
methods
. We introduced a quality index as a simple measure
of LPS purity in terms of a degree of polysaccharide content
detected by absorbance at 204 nm. Further experiments using
gel electrophoresis, endotoxin test, and macrophage activation
test were performed to evaluate the performance and reliability
of a proposed ‘T-sol’ method and the biological effectiveness
and character of the LPS products. We presented
that the T-sol method had differential effects on extraction of
a RAW 264.7 cell-activating LPS, which was effective in the
macrophage activation with similar effects in stimulating
the production of TNF-alpha. In conclusion, the T-sol method
provides a simple way to improve quality and biological activity
of LPS with high yield.

Abstract
-
The colonization of liquid surfaces as floating biofilms or pellicles
is a bacterial adaptation to optimally occupy the airliquid
(A-L) niche. In aerobic heterotrophs, pellicle formation
is beneficial for the utilization of O2 and nonpolar organic
compounds. Pseudomonas alkylphenolica KL28, an alkylphenol
degrader, forms flat circular pellicles that are 0.3–
0.5 mm in diameter. In this study, we first monitored the pellicle
developmental patterns of multicellular organization
from the initial settlement stage. The pellicles developed by
clonal growth and mutants for flagella and pilus formation
established normal pellicles. In contrast, the mutants of an
epm gene cluster for biosynthesis of alginate-like polymer
were incompetent in cell alignment for initial two-dimensional
(2D) pellicle growth, suggesting the role of the Epm
polymer as a structural scaffold for pellicle biofilms. Microscopic
observation revealed that the initial 2D growth transited
to multilayers by an accumulated self-produced extracellular
polymeric substance that may exert a constraint force.
Electron microscopy and confocal laser scanning microscopy
revealed that the fully matured pellicle structures were densly
packed with matrix-encased cells displaying distinct arrangements.
The cells on the surface of the pellicle were relatively
flat, and those inside were longitudinally cross-packed. The
extracellular polysaccharide stained by Congo red was denser
on the pellicle rim and a thin film was observed in the open
spaces, indicative of its role in pellicle flotation. Our results
demonstrate that P. alkylphenolica KL28 coordinately dictates
the cell arrangements of pellicle biofilms by the controlled
growth of constituent cells that accumulate extracellular
polymeric substances.

Nine Klebsiella pneumoniae isolates coproducing NDM-1
and OXA-232 carbapenemases were successively isolated
from a single patient. Although they were isolated simultaneously
and were isogenic, they presented different colony
phenotypes (matt and mucoid). All nine isolates were resistant
to most antibiotics except colistin and fosfomycin. In
addition, matt-type isolates were resistant to tigecycline. No
differences were detected in the cps cluster sequences, except
for the insertion of IS5 in the wzb gene of two matt-type isolates.
In vitro virulence assays based on production of capsular
polysaccharide, biofilm formation, and resistance to
human serum indicated that the mucoid-type isolates were
significantly more virulent than the matt-type. In addition,
mucoid-type isolates showed higher survival rates than the
matt-type ones in infection experiments in the fruit fly, suggesting
a higher virulence of K. pneumoniae isolates with a
mucoid phenotype. To our knowledge, this is the first report
of K. pneumoniae colonies with different phenotypes being
isolated from the same sample. In addition, we show that virulence
varies with colony phenotype. Dissemination of K.
pneumoniae isolates expressing both antibiotic resistance
and high virulence would constitute a great threat.

Abstract
-
Bacterial biofilms remain a persistent threat to human healthcare
due to their role in the development of antimicrobial
resistance. To combat multi-drug resistant pathogens, it is
crucial to enhance our understanding of not only the regulation
of biofilm formation, but also its contribution to bacterial
virulence. Iron acquisition lies at the crux of these two
subjects. In this review, we discuss the role of iron acquisition
in biofilm formation and how hosts impede this mechanism
to defend against pathogens. We also discuss recent findings
that suggest that biofilm formation can also have the reciprocal
effect, influencing siderophore production and iron
sequestration.

Abstract

Mammalian gut microbial communities form intricate mutualisms
with their hosts, which have profound implications
on overall health. One group of important gut microbial mutualists
are bacteria in the genus Ruminococcus, which serve
to degrade and convert complex polysaccharides into a variety
of nutrients for their hosts. Isolated decades ago from the bovine
rumen, ruminococci have since been cultured from other
ruminant and non-ruminant sources, and next-generation
sequencing has further shown their distribution to be widespread
in a diversity of animal hosts. While most ruminococci
that have been studied are those capable of degrading
cellulose, much less is known about non-cellulolytic, nonruminant-
associated species, such as those found in humans.
Furthermore, a mechanistic understanding of the role of
Ruminococcus spp. in their respective hosts is still a work in
progress. This review highlights the broad work done on
species within the genus Ruminococcus with respect to their
physiology, phylogenetic relatedness, and their potential impact
on host health.

It has been demonstrated that certain lactic acid bacteria (LAB) can sequester metal ions by binding them to their surfaces. In the present study, lead (Pb)-resistant LAB were isolated from kimchi, a Korean fermented food. A total of 96 different LAB strains were isolated, and 52 strains showed lead resistance. Among them, an LAB strain-96 (L-96) iden-tified as Leuconostoc mesenteroides showed remarkable Pb resistance and removal capacity. The maximum adsorption capacity of this strain calculated using the Langmuir isotherm was 60.6 mg Pb/g. In an in vivo experiment, young male mice were provided with water (A), Pb-water (B), or Pb-water+ L-96 (C) during puberty. Lower glutamate oxaloacetate trans-aminase (GOT) and glutamate pyruvate transaminase (GPT) levels in Pb-exposed male mice that received strain L-96 as a probiotic were suggestive of reduced hepatotoxicity. More-over, feces from mice treated with L-96 contained more Pb than feces from untreated mice. Increased Pb elimination likely reduced internal accumulation, and this hypothesis was supported by significantly lower Pb concentrations in kid-neys and testes of the mice treated with strain L-96. The mo-tility and ATP content of epididymal spermatozoa were par-tially restored if strain L-96 was administered. In conclusion, isolated L-96 LAB had lead-biosorption activity and effi-ciently detoxified lead-poisoned male mice, resulting in re-covering male reproductive function. These results suggest the potential use of LAB as a probiotic to protect humans from the adverse effects of Pb exposure.

Abstract

To gain insights into dimorphism, cell wall polysaccharides
from Tremella fuciformis strains were obtained from alkaliextracted
water-soluble fractions PTF-M38 (from the mycelial
form), PTF-Y3 and PTF-Y8 (from the yeast form) of
T. fuciformis strains were used to gain some insights into
dimorphism study. Their chemical properties and structural
features were investigated using gel permeation chromatography,
gas chromatography, UV and IR spectrophotometry
and Congo red binding reactions. The results indicated that
the backbones of PTF-M38, PTF-Y3 and PTF-Y8 were configured
with α-linkages with average molecular weights of
1.24, 1.08, and 1.19 kDa, respectively. PTF-M38 was mainly
composed of xylose, mannose, glucose, and galactose in a
ratio of 1:1.47:0.48:0.34, while PTF-Y3 and PTF-Y8 were
mainly composed of xylose, mannose and glucose in a ratio
of 1:1.65:4.06 and 1:1.21:0.44, respectively. The sugar profiles
of PTF-M38, PTF-Y3 and PTF-Y8 were also established
for further comparison. These profiles showed that all three
polysaccharides contained the same sugars but in different
ratios, and the carbon sources (xylose, mannose, glucose, and
galactose) affected the sugar ratios within the polysaccharides.

Abstract
-
Over the past several decades, research on the synthesis and
organization of the cell wall polysaccharides of Aspergillus
fumigatus has expanded our knowledge of this important
fungal structure. Besides protecting the fungus from environmental
stresses and maintaining structural integrity of
the organism, the cell wall is also the primary site for interaction
with host tissues during infection. Cell wall polysaccharides
are important ligands for the recognition of fungi
by the innate immune system and they can mediate potent
immunomodulatory effects. The synthesis of cell wall polysaccharides
is a complicated process that requires coordinated
regulation of many biosynthetic and metabolic pathways.
Continuous synthesis and remodeling of the polysaccharides
of the cell wall is essential for the survival of the
fungus during development, reproduction, colonization and
invasion. As these polysaccharides are absent from the human
host, these biosynthetic pathways are attractive targets
for antifungal development. In this review, we present recent
advances in our understanding of Aspergillus fumigatus cell
wall polysaccharides, including the emerging role of cell
wall polysaccharides in the host-pathogen interaction.
