With global expansion of the COVID-19 pandemic and the
emergence of new variants, extensive efforts have been made
to develop highly effective antiviral drugs and vaccines against
SARS-CoV-2. The interactions of coronaviruses with host
antiviral interferon pathways ultimately determine successful
viral replication and SARS-CoV-2-induced pathogenesis.
Innate immune receptors play an essential role in host defense
against SARS-CoV-2 via the induction of IFN production
and signaling. Here, we summarize the recent advances
in innate immune sensing mechanisms of SARS-CoV-2 and
various strategies by which SARS-CoV-2 antagonizes antiviral
innate immune signaling pathways, with a particular
focus on mechanisms utilized by multiple SARS-CoV-2 proteins
to evade interferon induction and signaling in host cell.
Understanding the underlying immune evasion mechanisms
of SARS-CoV-2 is essential for the improvement of vaccines
and therapeutic strategies.

RraA, a protein regulator of RNase E activity, plays a unique
role in modulating the mRNA abundance in Escherichia coli.
The marine pathogenic bacterium Vibrio vulnificus also possesses
homologs of RNase E (VvRNase E) and RraA (VvRraA1
and VvRraA2). However, their physiological roles have not
yet been investigated. In this study, we demonstrated that
VvRraA1 expression levels affect the pathogenicity of V. vulnificus.
Compared to the wild-type strain, the VvrraA1-deleted
strain (ΔVvrraA1) showed decreased motility, invasiveness,
biofilm formation ability as well as virulence in mice; these
phenotypic changes of ΔVvrraA1 were restored by the exogenous
expression of VvrraA1. Transcriptomic analysis indicated
that VvRraA1 expression levels affect the abundance
of a large number of mRNA species. Among them, the halflives
of mRNA species encoding virulence factors (e.g., smcR
and htpG) that have been previously shown to affect VvrraA1
expression-dependent phenotypes were positively correlated
with VvrraA1 expression levels. These findings suggest that
VvRraA1 modulates the pathogenicity of V. vulnificus by regulating
the abundance of a subset of mRNA species.

Thyroid carcinoma is a common endocrine organ cancer associated
with abnormal hormone secretion, leading to the
disorder of metabolism. The intestinal microbiota is vital to
maintain digestive and immunologic homeostasis. The relevant
information of the microbial community in the gut and
thyroid, including composition, structure, and relationship,
is unclear in thyroid carcinoma patients. A total of 93 samples
from 25 patients were included in this study. The results
showed that microbial communities existed in thyroid tissue;
gut and thyroid had high abundance of facultative anaerobes
from the Proteobacteria phyla. The microbial metabolism from
the thyroid and gut may be affected by the thyroid carcinoma
cells. The cooccurrence network showed that the margins of
different thyroid tissues were unique areas with more competition;
the stabilization of microcommunities from tissue
and stool may be maintained by several clusters of species
that may execute different vital metabolism processes dominantly
that are attributed to the microenvironment of cancer.

We previously reported that human cytomegalovirus (HCMV)
86 kDa immediate-early 2 gene product (IE86) promotes
proteasome-dependent degradation of STING. In the present
study, we determined the specific residues of IE86 responsible
for STING degradation using a STING-firefly luciferase
fusion protein expression system for quantitative measurement
of STING protein levels. IE86 amino acids (aa)
136–289 were sufficient to promote STING degradation and
further induced down-regulation of 23-cyclic GMP-AMP
(cGAMP)-mediated IFN-β promoter activation. Interestingly,
transactivation domains (TAD) of the IE86 protein located
at the N- and C-termini were required for down-regulation
of Toll/interleukin-1 receptor (TIR) domain-containing adaptor-
inducing interferon β (IFN-β) (TRIF)-mediated IFN-β-
and p65/RelA-induced NF-κB-dependent promoter activation
while amino acids (aa) 136–289 had no significant effects.
Our collective data suggest that the IE86 protein utilizes the
aa 136–289 region to promote STING degradation and inhibit
the cGAS-STING pathway.

The growing use of immunosuppressive therapies has resulted
in a dramatic increased incidence of invasive fungal
infections (IFIs) caused by Aspergillus fumigatus, a common
pathogen, and is also associated with a high mortality rate.
Azoles are the primary guideline-recommended therapy agents
for first-line treatment and prevention of IFIs. However,
increased azole usage in medicinal and agricultural settings
has caused azole-resistant isolates to repeatedly emerge in
the environment, resulting in a significant threat to human
health. In this review, we present and summarize current
research on the resistance mechanisms of azoles in A. fumigatus
as well as efficient susceptibility testing methods. Moreover,
we analyze and discuss the putative clinical (bedside)
indication of these findings from bench work.

Loop-mediated isothermal amplification (LAMP), a newly developed gene amplification method, combines rapidity, simplicity, and high specificity. Several tests have been developed based on this method, and simplicity is maintained throughout all steps, from extraction of nucleic acids to detection of amplification. In the LAMP reaction, samples are amplified at a fixed temperature through a repetition of two types of elongation reactions occurring at the loop regions: self-elongation of templates from the stem loop structure formed at the 3'-terminal and the binding and elongation of new primers to the loop region. The LAMP reaction has a wide range of possible applications, including point-of-care testing, genetic testing in resource-poor settings (such as in developing countries), and rapid testing of food products and environmental samples.
